Control of protein adsorption and release by stimuli-responsive polymer brushes

Thin polymer brushes have proven their ability to modify physico-chemical interface properties as well as the adsorption behaviour of proteins at artificial surfaces. Especially brushes, that are highly swellable in aqueous solution, are promising for drug delivery or sensor applications. Here it is necessary to understand the principles of swelling and its influence on the adsorption behavior in order to perform controlled stimuli-responsive adsorption and release of biomolecules. With the help of in-situ spectroscopic ellipsometry these processes can be monitored quantitatively providing informations for a better understanding of the complex behavior of this combined polymer – protein interfaces. We report on swelling measurements of homopolymer and mixed polymer brushes consisting of pH- and temperature sensitive polymer chains. Furthermore we investigated the adsorption of different proteins onto charged polymer brushes. Here we monitored the influence of changes in the environmental conditions on the adsorbed amount as well as on the layer properties of the formed mixed brush – protein phase and tested the reversibility of adsorption.
